On June 25, 2020, Javier Cortés published in the medical journal Journal of Clinical Oncology the "Efficacy and safety validation results from the ZETA Phase 3 clinical trial subgroup analysis of VEGFR/EGFR kinase inhibitor vandetanib in progressive and symptomatic medullary thyroid cancer patients."
The ZETA Phase 3 clinical trial randomized progressive/symptomatic medullary thyroid cancer patients (N=331) in a 2:1 ratio into the following two groups, with the primary endpoint being progression-free survival and secondary endpoints being objective response rate and overall survival.
- Vandetanib group (once-daily vandetanib 300 mg, N=231).
- Placebo group (N=100).
The trial included four patient categories: 1. Progressive and symptomatic; 2. Symptomatic only; 3. Progressive only; 4. Neither progressive nor symptomatic. The subgroup analysis target was progressive and symptomatic patients (N=184).
Subgroup analysis results: the primary endpoint median progression-free survival for progressive and symptomatic medullary thyroid cancer patients was: vandetanib group 21.43 months / placebo group 8.40 months.
Regarding safety, the adverse events that occurred in the vandetanib group were consistent with those seen in other clinical trials. The most commonly observed adverse events included digestive system issues and skin problems.
Based on the ZETA clinical trial subgroup analysis results, Michael C. Kreissl concluded: In progressive and symptomatic medullary thyroid cancer patients, VEGFR/EGFR kinase inhibitor vandetanib improved progression-free survival with clinically significant effect.
