On August 4, 2023, Eli Lilly announced the results of the Phase 3 LIBRETTO-431 clinical trial evaluating first-line Retevmo (selpercatinib) monotherapy in patients with RET fusion-positive metastatic or advanced non-small cell lung cancer (NSCLC).

The LIBRETTO-431 Phase 3 trial randomized 261 treatment-naive patients with RET fusion-positive metastatic or advanced NSCLC to receive either Retevmo monotherapy or anti-PD-1 antibody therapy combined with chemotherapy. The primary endpoint was progression-free survival (PFS), and secondary endpoints included overall survival (OS), objective response rate (ORR), duration of response (DOR), and intracranial objective response rate.

This trial is the first to compare a targeted therapy against anti-PD-1 therapy plus chemotherapy in patients with a specific genetic alteration. The results showed that Retevmo monotherapy significantly prolonged PFS compared to the control group. Detailed findings will be presented at an upcoming medical conference and published in a peer-reviewed journal.
