On August 9, 2022, Daiichi Sankyo announced the initiation of patient dosing in the HERTHENA-Lung02 Phase III clinical trial evaluating Patritumab Deruxtecan as second-line treatment for EGFR-mutant metastatic non-small cell lung cancer patients.

Lung cancer is the second most common cancer worldwide. Non-small cell lung cancer accounts for 80-85% of lung cancer cases, of which approximately 30% harbor EGFR mutations. With the advent of molecular targeted drugs and immune checkpoint inhibitors, EGFR tyrosine kinase inhibitors have been established as treatment options for EGFR-mutant advanced non-small cell lung cancer. However, most patients receiving EGFR tyrosine kinase inhibitor therapy eventually develop resistance, creating a need for new treatment options.

The HERTHENA-Lung02 Phase III clinical trial will treat EGFR-mutant metastatic non-small cell lung cancer patients who have received prior EGFR tyrosine kinase inhibitor therapy with patritumab deruxtecan and evaluate its efficacy and safety. The trial is currently being conducted in Japan, Europe, North America, and Oceania, with an expected enrollment of 560 patients.

The primary efficacy endpoint is progression-free survival, and secondary endpoints include overall survival, objective response rate, clinical benefit rate, disease control rate, and duration of response.

Daiichi Sankyo stated in its press release that it will accelerate the development of this drug to provide a new treatment option for EGFR-mutant metastatic non-small cell lung cancer patients.
