From May 29–31, 2020, Alan Loh Ho presented at the ASCO 2020 Virtual Meeting the results of "Efficacy and safety of farnesyltransferase inhibitor Tipifarnib monotherapy in patients with HRAS-mutant head and neck squamous cell carcinoma, thyroid cancer, salivary gland tumors, and urothelial carcinoma in the KO-TIP-001 and IST-01 Phase 2 clinical trials."

This Phase 2 clinical trial administered Tipifarnib monotherapy (28-day cycle, Days 1–7 and 15–21, twice-daily Tipifarnib 600–900 mg) to patients with HRAS-mutant head and neck squamous cell carcinoma, thyroid cancer, salivary gland tumors, and urothelial carcinoma. Primary endpoints were objective response rate and 6-month progression-free survival.

In the KO-TIP-001 and IST-01 trials, among evaluable head and neck squamous cell carcinoma patients (N=18): objective response rate was 56% (N=10/18), median progression-free survival was 6.1 months. Among recurrent and metastatic salivary gland tumor patients (N=12): objective response rate was 8% (N=1), median progression-free survival was 7 months. Among urothelial carcinoma patients (N=12): objective response rate was 42% (N=5), median progression-free survival was 5.1 months.

Based on the results of the KO-TIP-001 and IST-01 trials, Alan Loh Ho concluded: In patients with HRAS-mutant head and neck squamous cell carcinoma, thyroid cancer, salivary gland tumors, and urothelial carcinoma, the farnesyltransferase inhibitor Tipifarnib demonstrated anticancer activity.
