On January 19, 2023, Jazz Pharmaceuticals and Zymeworks announced initial analysis results from a Phase II clinical trial evaluating zanidatamab plus chemotherapy as first-line treatment in HER2-positive metastatic gastroesophageal adenocarcinoma patients.

This Phase II trial enrolled HER2-positive metastatic gastroesophageal adenocarcinoma patients (N=42) and administered zanidatamab plus chemotherapy as first-line treatment. Evaluation endpoints included overall survival (OS), objective response rate (ORR), and disease control rate (DCR).

Initial Analysis Results at Median Follow-up of 26.5 Months:

  • Median OS: not reached
  • 18-month OS rate: 84%
  • 12-month OS rate: 88%
  • Objective response rate: 79%
  • Disease control rate: 92%
  • Median duration of response: 20.4 months
  • Median progression-free survival: 12.5 months

Safety:

Adverse events observed in this trial were consistent with those seen in prior clinical trials and were within manageable range.

Based on these initial analysis results, lead investigator Dr. Elena Elimova of Princess Margaret Cancer Centre stated that gastroesophageal adenocarcinoma carries a high cancer mortality rate, and compared with OS data from previous clinical trials, zanidatamab plus chemotherapy demonstrated exceptionally favorable outcomes in this trial.
