FLOT Therapy as Perioperative Chemotherapy Improves Overall Survival in Resectable Gastric or Gastroesophageal Junction Adenocarcinoma
On April 12, 2019, the medical journal The Lancet published a Phase II/III clinical trial evaluating FLOT (docetaxel + oxaliplatin + leucovorin + fluorouracil) therapy as perioperative chemotherapy in patients with resectable gastric or gastroesophageal junction (GEJ) adenocarcinoma.
Patients received perioperative chemotherapy comparing FLOT therapy (docetaxel + oxaliplatin + fluorouracil + leucovorin) versus ECF/ECX therapy (epirubicin + cisplatin + fluorouracil or capecitabine) for efficacy and safety.
In overall survival statistics, FLOT therapy showed a statistically significant improvement in OS. Safety-wise, the differences between the two regimens were not large. Although FLOT therapy had a mortality risk of 23%, it improved patients' overall survival more than ECF/ECX therapy.
