On June 6, 2022, Karen L. Reckamp et al. from Cedars-Sinai Medical Center published results in the Journal of Clinical Oncology on the efficacy and safety of Ramucirumab + anti-PD-1 antibody in the Lung-MAP S1800A Phase II clinical trial for previously treated advanced non-small cell lung cancer patients with prior immune checkpoint inhibitor experience.
The Lung-MAP S1800A Phase II clinical trial randomized previously treated advanced non-small cell lung cancer patients with prior immune checkpoint inhibitor experience (N=136) into the Ramucirumab + anti-PD-1 antibody group (N=69) and the standard chemotherapy group (N=67). The primary endpoint was overall survival, and secondary endpoints included objective response rate and duration of response.
The trial results showed that the primary endpoint median overall survival was 14.5 months for the Ramucirumab + anti-PD-1 antibody group / 11.6 months for the standard chemotherapy group, with the Ramucirumab + anti-PD-1 antibody group showing a 31% reduction in death risk. The death risk for progression-free survival was reduced by 14%. The objective response rate was 22% for the Ramucirumab + anti-PD-1 antibody group / 28% for the standard chemotherapy group.
Regarding safety, the incidence of grade 3 or higher adverse events was 42% for the Ramucirumab + anti-PD-1 antibody group / 60% for the standard chemotherapy group.
Based on the Lung-MAP S1800A trial results, Karen L. Reckamp et al. stated that Ramucirumab + anti-PD-1 antibody therapy demonstrated improved overall survival compared with standard chemotherapy in previously treated advanced non-small cell lung cancer patients with prior immune checkpoint inhibitor experience, with a safety profile consistent with known data.
