On September 10, 2019, Bristol-Myers Squibb announced pooled five-year follow-up analysis results for previously treated advanced non-small cell lung cancer patients receiving anti-PD-1 antibody anti-PD-1 antibody monotherapy.

This pooled analysis was based on the Phase III clinical trials CheckMate-017 and CheckMate-057. Both trials enrolled non-small cell lung cancer patients who had progressed after first-line treatment. Patients were randomized 1:1 into the anti-PD-1 antibody monotherapy group (3 mg/kg every two weeks) and the docetaxel monotherapy group (75 mg/m2 every three weeks). The primary endpoint was overall survival. Additionally, the CheckMate-017 trial enrolled squamous cell carcinoma patients, while the CheckMate-057 trial enrolled non-squamous cell carcinoma patients.

The pooled analysis results showed that the primary endpoint 5-year overall survival rate was 13.4% for the anti-PD-1 antibody group / 2.6% for the docetaxel group. Subgroup analysis reported higher 5-year survival rates in the anti-PD-1 antibody group across the subgroups examined (literature data; for reference only).

For secondary endpoints, the objective response rate showed that approximately 32.2% of patients who responded to anti-PD-1 antibody maintained their response at year 5 / no patients in the docetaxel group maintained response at year 5. The median duration of response was 19.9 months / 5.6 months, respectively. Regarding safety, the adverse events observed in this analysis were consistent with the known safety profile from other clinical trials, with no new adverse events identified.

Based on this pooled analysis, Scott Gettinger concluded that previously treated advanced non-small cell lung cancer patients receiving anti-PD-1 antibody anti-PD-1 antibody monotherapy demonstrated significantly greater benefit compared to conventional chemotherapy, even with long-term treatment.
