On September 11, 2023, Daiichi Sankyo presented results at the World Conference on Lung Cancer held in Singapore on the efficacy and safety of anti-HER2 antibody-drug conjugate in the DESTINY-Lung02 Phase II clinical trial for HER2-mutant advanced non-small cell lung cancer.
This trial enrolled previously treated HER2-mutant advanced non-small cell lung cancer patients and administered anti-HER2 antibody-drug conjugate at 5.4 mg/kg (N=102) and 6.4 mg/kg (N=50). The primary endpoint was objective response rate, and secondary endpoints included duration of response and overall survival.
The trial results showed that the primary endpoint objective response rate was: 49.0% for the 5.4 mg/kg group (1 complete response, 49 partial responses) and 56.0% for the 6.4 mg/kg group (2 complete responses, 26 partial responses).
The secondary endpoint median duration of response was: 16.8 months for the 5.4 mg/kg group / not reached for the 6.4 mg/kg group. The median progression-free survival was: 9.9 months for the 5.4 mg/kg group / 15.4 months for the 6.4 mg/kg group. The median overall survival was: 19.5 months for the 5.4 mg/kg group / not reached for the 6.4 mg/kg group.
Regarding safety, grade 3 or higher adverse events included neutropenia (5.4 mg/kg group 18.8% / 6.4 mg/kg group 36.0%) and anemia (5.4 mg/kg group 10.9% / 6.4 mg/kg group 16.0%). Although drug-related interstitial lung disease was observed (5.4 mg/kg group 12.9% / 6.4 mg/kg group 28.0%), most were low-grade adverse events. The grade 3 adverse event rates were: 5.4 mg/kg group 1.0% / 6.4 mg/kg group 0%, and grade 5 adverse event rates were: 5.4 mg/kg group 1.0% / 6.4 mg/kg group 2.0%.
