On January 24, 2022, Julien Mazieres et al. from Universite Paul Sabatier published in the medical journal Journal of Clinical Oncology the efficacy and safety results of Trastuzumab + Pertuzumab + Docetaxel for previously treated HER2-positive non-small cell lung cancer patients in the IFCT-1703 R2D2 Phase II clinical trial.
The IFCT-1703 R2D2 Phase II clinical trial was a multi-center randomized trial conducted in three-week treatment cycles, administering Trastuzumab 6 mg/kg (initial dose 8 mg/kg) + Pertuzumab 420 mg (initial dose 840 mg) + Docetaxel 75 mg/m2 to HER2-positive non-small cell lung cancer patients with prior platinum-based therapy (N=45). The primary endpoint was objective response rate, with secondary endpoints including duration of treatment, progression-free survival, and safety.
Patient characteristics included a median age of 64.5 years (31-84), 35% smokers, 72% female, 15% with performance status score of 2, and 30% with brain metastases. The study results were as follows:
The primary endpoint objective response rate was 29% (N=13), with a disease stabilization rate of 58% (N=26). The secondary endpoint median progression-free survival was 6.8 months, and the median duration of response was 11 months.
Regarding safety, the grade 3-4 adverse event rate was 64%, with a treatment discontinuation rate due to adverse events of 0%. The most common adverse events were neutropenia 33%, diarrhea 13%, and anemia 9%.
Based on the IFCT-1703 R2D2 trial results, Julien Mazieres et al. stated that Trastuzumab + Pertuzumab + Docetaxel for previously treated HER2-positive non-small cell lung cancer patients demonstrated good anti-tumor efficacy with acceptable tolerability, and this regimen should be considered as a treatment option for these patients in the future.
