On March 9, 2023, AstraZeneca announced results from the AEGEAN Phase III clinical trial evaluating the efficacy and safety of Imfinzi + chemotherapy as perioperative treatment for patients with resectable stage IIA-IIIB non-small cell lung cancer.
The AEGEAN clinical trial was a multicenter, double-blind Phase III clinical trial that randomized patients with resectable stage IIA-IIIB non-small cell lung cancer (N=802) into an Imfinzi (1500 mg) + chemotherapy group (neoadjuvant therapy: four cycles of three-week treatment cycles; adjuvant therapy: Imfinzi 1500 mg every four weeks for up to 12 cycles) and a placebo group. The primary endpoints were pathological complete response rate after adjuvant therapy and event-free survival, with overall survival, safety, and quality of life as additional endpoints.
The trial results showed that the Imfinzi + chemotherapy group demonstrated significant improvement in event-free survival compared with the placebo group. The other primary endpoint of pathological complete response also showed favorable results consistent with previous reports. Regarding safety, the findings were consistent with the known safety profile of Imfinzi from existing clinical trials, and no new adverse events were identified in this study.
Based on the AEGEAN Phase III clinical trial results, Susan Galbraith, AstraZeneca Executive Vice President, stated that patients with resectable non-small cell lung cancer face a high risk of recurrence despite receiving chemotherapy and surgery. Using Imfinzi before and after surgery may extend the time patients can live normally without recurrence or progression, and overall survival will continue to be monitored.
