For patients with early-stage liver cancer, surgical resection or radiofrequency ablation offers hope for a cure. However, the accompanying high risk of recurrence (over 70%) has long been a shared concern for both doctors and patients. In January 2023, Roche Pharmaceuticals published interim analysis results from the IMbrave050 Phase 3 clinical trial, bringing a new possibility for safeguarding the health of these patients.
IMbrave050: The First Phase 3 Trial to Confirm Benefits of Adjuvant Therapy
This study targeted patients with early-stage hepatocellular carcinoma (HCC) who are at high risk of recurrence after radical resection or radiofrequency ablation. The trial randomized 662 patients into two groups to verify the protective capacity of this immunotherapy combination:
- Tecentriq + Avastin Group: Tecentriq (1200mg) combined with Avastin (15 mg/kg), every three weeks for up to 12 months.
- Active Surveillance Group.
The primary objective of the study was to observe Recurrence-Free Survival (RFS) as assessed by an independent review facility.
Encouraging Interim Results: Significantly Reducing Recurrence
The interim analysis showed that Tecentriq + Avastin adjuvant therapy significantly improved Recurrence-Free Survival compared to active surveillance alone. This means the combination therapy can effectively delay or even prevent the return of the tumor, adding a solid layer of protection to the post-surgical recovery journey.
While Overall Survival (OS) requires longer-term follow-up for a final conclusion, the preliminary success shown thus far is highly encouraging.
Consistent Safety Profile
Regarding safety, the performance of Tecentriq combined with Avastin was consistent with its known profile in advanced liver cancer treatment, with no new safety signals identified. This stability gives medical teams greater confidence in formulating post-surgical management plans and allows patients to maintain a good quality of life during treatment.
Dr. Levi Garraway, Roche’s Chief Medical Officer, noted that IMbrave050 is the first Phase 3 trial to achieve success in the adjuvant setting for liver cancer—a advance that represents both a medical milestone and a warm promise to countless families.
