The EMPOWER-Cervical 1/GOG-3016/ENGOT-cx9 Phase 3 clinical trial evaluated cemiplimab (anti-PD-1 antibody) versus investigator's choice of chemotherapy in patients with recurrent cervical cancer who had progressed on prior platinum-based therapy. Results were published in the New England Journal of Medicine.
Trial Design:
Patients with recurrent, metastatic, or persistent cervical cancer who had progressed on prior platinum-containing chemotherapy were randomized to receive either cemiplimab 350 mg IV every 3 weeks or investigator's choice of single-agent chemotherapy (topotecan, vinorelbine, gemcitabine, irinotecan, or pemetrexed). The primary endpoint was overall survival (OS).
Results:
- Median OS: Cemiplimab 12.0 months vs. chemotherapy 8.5 months
- Hazard ratio: 0.69 (31% reduction in death risk)
- The OS benefit was observed regardless of PD-L1 expression status
Conclusion: Based on the EMPOWER-Cervical 1 trial results, cemiplimab demonstrated a statistically significant and clinically meaningful improvement in overall survival compared to chemotherapy in patients with recurrent cervical cancer following platinum-based therapy.
