On October 5, 2021, Dr. Hongyan Cheng and colleagues from Peking Union Medical College Hospital published results in The Lancet from a Phase 2 clinical trial evaluating the efficacy and safety of the anti-PD-1 antibody camrelizumab combined with apatinib in patients with high-risk, chemotherapy-resistant, recurrent gestational trophoblastic tumors (GTT).
This Phase 2 trial was a single-arm, open-label study with a 4-week treatment cycle. Patients with high-risk, chemotherapy-resistant, recurrent GTT received camrelizumab (200 mg on days 1 and 15) plus apatinib (250 mg once daily) until disease progression or unacceptable toxicity. The primary endpoint was objective response rate (ORR).
Results at median follow-up of 18.5 months:
- Objective response rate (ORR): 55%
- Complete response (CR): 50%
Safety:
Grade 3 or higher adverse events observed in most patients included hypertension (25%, N=5), skin abnormalities (20%, N=4), and neutropenia (10%, N=2). No higher-grade adverse events were confirmed.
Conclusion: Based on the Phase 2 trial results, Dr. Cheng and colleagues concluded that camrelizumab plus apatinib demonstrated meaningful anti-tumor activity with acceptable tolerability in high-risk, chemotherapy-resistant, recurrent GTT patients, and may represent a new treatment option for this patient population.
