On July 24, 2019, Roche announced that the Phase 3 IMvigor130 clinical trial met its co-primary endpoint of progression-free survival (PFS). The study evaluated Atezolizumab (Tecentriq) in combination with platinum-based chemotherapy for patients with previously untreated locally advanced or metastatic urothelial carcinoma (bladder cancer).
Study Design: IMvigor130 Trial
IMvigor130 is an international, multicenter, randomized, double-blind Phase 3 trial. It enrolled 1,213 patients who were randomized into three treatment arms:
- Atezolizumab + Chemotherapy (Gemcitabine plus Cisplatin or Carboplatin).
- Atezolizumab Monotherapy.
- Placebo + Chemotherapy.
The primary endpoints were PFS and overall survival (OS).
Key Results
- Progression-Free Survival (PFS): The combination of Atezolizumab and chemotherapy demonstrated a statistically significant improvement in PFS compared to chemotherapy alone.
- Overall Survival (OS): At the time of the initial report, OS data were not yet mature, and follow-up was ongoing.
- Safety: The safety profile of the Atezolizumab combination was consistent with the known profiles of the individual medicines, with no new safety signals observed.
Expert Commentary
Dr. Sandra Horning, Chief Medical Officer at Roche, stated that IMvigor130 is the first Phase 3 study to demonstrate that an immune checkpoint inhibitor combined with chemotherapy can improve outcomes in the first-line treatment of advanced bladder cancer.
