Tecentriq + Cobimetinib Combination Therapy Failed to Extend Overall Survival in Previously Treated Locally Advanced or Metastatic Colorectal Cancer Patients

On April 16, 2019, The Lancet published comparative Phase 3 clinical trial efficacy results for previously treated locally advanced or metastatic colorectal cancer patients using Tecentriq (atezolizumab) + cobimetinib combination therapy, Tecentriq monotherapy, and regorafenib monotherapy.

This trial enrolled patients who had received at least two prior lines of treatment. The primary endpoint was overall survival, and secondary endpoints were progression-free survival, objective response rate, and duration of response. The most commonly occurring adverse events from this combination therapy and their rates were: diarrhea 11%, anemia 6%, elevated creatine phosphokinase 7%, and fatigue 4%.

Based on the trial results, both the combination therapy and Tecentriq monotherapy did not show significant improvement in overall survival compared to regorafenib monotherapy.
