AGO-OVER 17 BOOST Phase III Trial Results

On November 4, 2022, researchers led by Jacobus Pfisterer from Arbeitsgemeinschaft Gynaekologische Onkologie published AGO-OVER 17 BOOST/GINECO OV118/ENGOT Ov-15 Phase III results in the Journal of Clinical Oncology, evaluating extended bevacizumab duration in first-line ovarian cancer treatment.
Trial Design
Patient Population: Stage IIB-IV advanced epithelial ovarian cancer (n=927)
Treatment Arms:
- Paclitaxel 175 mg/m² + Carboplatin + Bevacizumab 15 mg/kg for 15 months (n=469)
- Paclitaxel 175 mg/m² + Carboplatin + Bevacizumab 15 mg/kg for 30 months (n=458)
Primary Endpoint: Progression-free survival
Key Results
Progression-Free Survival Median:
- 15-month bevacizumab: 24.2 months
- 30-month bevacizumab: 26.0 months
- No significant difference
Overall Survival: No significant difference between groups
Safety Profile
Grade 3-4 adverse events: 29% (15-month group) vs 34% (30-month group), consistent with established safety profile.
Clinical Conclusions
Extended bevacizumab beyond 15 months does not provide additional progression-free or overall survival benefit in first-line advanced ovarian cancer treatment.
