(Prostate Cancer) Is PARP Inhibitor Rucaparib Effective?

On August 14, 2020, Wassim Abida published results in the Journal of Clinical Oncology from the TRITON2 Phase 2 clinical trial evaluating the efficacy and safety of the PARP inhibitor Rucaparib in patients with BRCA1/BRCA2 gene mutation-positive, metastatic castration-resistant prostate cancer.
The TRITON2 Phase 2 trial enrolled 115 previously treated patients with BRCA1/BRCA2 mutation-positive metastatic castration-resistant prostate cancer, who received Rucaparib monotherapy twice daily. The primary endpoints were objective response rate (ORR) and PSA response rate.
Approximately 12% of metastatic castration-resistant prostate cancer patients carry BRCA1 or BRCA2 gene mutations. These mutations increase the risk of prostate cancer development and metastasis. Because PARP inhibitors can suppress homologous recombination repair defects and DNA damage responses caused by BRCA1/BRCA2 mutations, they were expected to benefit this patient population.
Results:
- Primary endpoint ORR (IRC-assessed): 43.5%
- Primary endpoint ORR (investigator-assessed): 50.8%
- PSA response rate: 54.8%
Conclusion: Based on the TRITON2 trial results, Wassim Abida concluded that Rucaparib demonstrated meaningful anti-tumor activity in BRCA1/BRCA2 mutation-positive metastatic castration-resistant prostate cancer patients, with a safety profile consistent with previously known adverse event data from other clinical trials.
