On December 9, 2022, AstraZeneca presented at the 2022 San Antonio Breast Cancer Symposium the efficacy and safety results of datopotamab deruxtecan in previously treated, metastatic triple-negative breast cancer (TNBC) from the TROPION-PanTumor01 Phase 1 trial.
Trial Design
TROPION-PanTumor01 was a Phase 1 trial administering datopotamab deruxtecan 6 mg/kg or 8 mg/kg (every 3 weeks) to previously treated patients with metastatic TNBC (N=44). Primary endpoint: Safety. Secondary endpoint: Objective response rate (ORR).
Efficacy Results
Overall population:
- ORR: 32% (complete response: 1, partial response: 13, stable disease: 18)
- Median PFS: 4.4 months
- Median OS: 13.5 months
- Disease control rate: 80%
Subgroup — Topoisomerase inhibitor-naive patients (N=27):
- ORR: 44% (complete response: 1, partial response: 11, stable disease: 10)
- Median PFS: 7.3 months
- Median OS: 14.3 months
Safety Profile
Datopotamab deruxtecan safety profile was consistent with current known data. Grade 3+ adverse events:
- Stomatitis: 11%
- Lymphopenia: 7%
- Fatigue: 7%
- Vomiting: 5%
- Anemia: 2%
- Neutropenia: 2%
- Nausea: 2%
Serious adverse event rate: 20.5%. One patient discontinued treatment due to pneumonitis. No febrile neutropenia or diarrhea was confirmed in patients.
Conclusions
Based on TROPION-PanTumor01 results, Aditya Bardia et al. from Mass General Cancer Center concluded that TNBC is the most aggressive form of breast cancer with OS under 18 months. In previously treated, metastatic TNBC patients, datopotamab deruxtecan demonstrated sustained and promising antitumor activity.
