TIL Therapy Combined with Nivolumab in Anti-PD-1 Resistant NSCLC

On August 12, 2021, Benjamin C. Creelan and colleagues at H. Lee Moffitt Cancer Center & Research Institute published in the medical journal Nature Medicine the efficacy and safety results of a Phase 1 clinical trial evaluating tumor-infiltrating lymphocyte (TIL) therapy combined with the anti-PD-1 antibody nivolumab in patients with advanced non-small cell lung cancer (NSCLC) resistant to anti-PD-1 therapy.
This single-arm Phase 1 trial enrolled patients with anti-PD-1 antibody-resistant NSCLC who had experienced disease progression after first-line nivolumab treatment. Patients received TIL therapy combined with nivolumab. The primary endpoint was safety, and the secondary endpoint was objective response rate (ORR).
Safety results (primary endpoint): The incidence of serious adverse events with TIL therapy plus nivolumab was 17% or below.
Efficacy results (secondary endpoint): Among 13 evaluable patients, 3 patients responded to treatment, tumor shrinkage was confirmed in 11 patients, and 2 achieved complete response, with a duration of 1.5 years.
Based on these Phase 1 results, Benjamin C. Creelan and colleagues concluded that TIL therapy combined with nivolumab was well tolerated in patients with anti-PD-1 resistant advanced NSCLC and may represent a promising new treatment option.
