From September 27 to October 1, 2019, at the European Society for Medical Oncology conference in Barcelona, Lori J. Wirth presented "Efficacy, Safety, and RET Mutation-Positive Thyroid Cancer Cohort Analysis Results of Oral RET Inhibitor Selpercatinib (LOXO-292) Monotherapy in Previously Treated Solid Tumor Patients from the LIBRETTO-001 Phase 1/2 Trial."
The LIBRETTO-001 Phase 1/2 trial evaluates selpercatinib (LOXO-292) monotherapy in solid tumor patients with prior multiple treatments. The primary endpoint is objective response rate, with secondary endpoints including duration of response, progression-free survival, and overall survival.
The cohort analysis results for RET mutation-positive thyroid and medullary thyroid cancer were: thyroid cancer patients (N=26) achieved a 62% objective response rate as the primary endpoint; medullary thyroid cancer patients (N=226) achieved a 56% objective response rate as the primary endpoint.
Regarding safety, adverse events confirmed in ≥15% of patients (including other solid tumor patients) included thirst, diarrhea, hypertension, ALT elevation, and AST elevation—most were Grade 1 or 2 adverse events. The discontinuation rate due to adverse events was 1.7% (N=9 patients).
Based on the LIBRETTO-001 trial results, Lori J. Wirth concluded: Previously treated RET mutation-positive thyroid cancer and medullary thyroid cancer patients treated with selpercatinib (LOXO-292) monotherapy demonstrate good objective response rates with acceptable tolerability to adverse events.
