(Ovarian Cancer) Is Fuzuloparib Effective?

On April 11, 2022, Ning Li et al. from the National Cancer Center published results from the FZOCUS-2 Phase III clinical trial in the Journal of Clinical Oncology, evaluating the efficacy and safety of PARP inhibitor Fuzuloparib as maintenance therapy in platinum-sensitive recurrent ovarian cancer patients with two or more prior treatments.
The FZOCUS-2 Phase III trial was a multicenter, double-blind, controlled trial that randomly assigned platinum-sensitive recurrent ovarian cancer patients with two or more prior treatments (N=252) in a 2:1 ratio to either the Fuzuloparib group (150mg twice daily, N=167) or the placebo group (N=85). The primary endpoint was progression-free survival (PFS), and the secondary endpoint was PFS in patients with BRCA1/2 mutations.
The trial results showed that the median PFS (primary endpoint) demonstrated improvement in the Fuzuloparib group compared to the placebo group. The secondary endpoint of PFS in BRCA1/2 mutation-positive patients also showed improvement in the Fuzuloparib group compared to the placebo group. Additionally, in BRCA1/2 wild-type patients, PFS also showed improvement in the Fuzuloparib group compared to the placebo group.
Regarding safety, the grade 3 or higher adverse events confirmed in the Fuzuloparib group were anemia (25.1%), thrombocytopenia (16.8%), and neutropenia (12.6%). One patient (0.6%) discontinued treatment due to adverse events.
Based on the FZOCUS-2 trial results, Ning Li et al. stated: "PARP inhibitor Fuzuloparib as maintenance therapy improved progression-free survival in platinum-sensitive recurrent ovarian cancer patients with two or more prior treatments, regardless of BRCA1/2 mutation status, with manageable adverse events."
