Is Tecentriq + Carboplatin + Pemetrexed Effective for Lung Cancer?

On August 21, 2023, Ernest Nadal and colleagues from L’Hospitalet de Llobregat published the results of a Phase 2 clinical trial in the "Journal of Clinical Oncology." The study evaluated the efficacy and safety of Tecentriq (atezolizumab) + Carboplatin + Pemetrexed in treatment-naive patients with advanced non-squamous non-small cell lung cancer (NSCLC) and brain metastases.

In this Phase 2 trial, 42 treatment-naive patients with advanced non-squamous NSCLC and brain metastases received Tecentriq + Carboplatin + Pemetrexed (for 4-6 cycles, followed by maintenance Tecentriq + Pemetrexed for up to 2 years or until disease progression). The primary endpoints were the 12-week progression-free survival (PFS) rate and the incidence of Grade 3 or higher adverse events from the start of treatment through week 9.

The 12-week PFS rate was 62.2%, and the incidence of Grade 3 or higher adverse events through week 9 was 27.5%. Most neurological adverse events were Grade 1-2, with Grade 3-4 events occurring in 12.5% (N=5) of patients.

Other endpoints included a median intracranial PFS of 6.9 months, an intracranial response rate of 42.7%, a median overall survival (OS) of 11.8 months, and a 2-year OS rate of 27.5%.

Based on these Phase 2 results, Ernest Nadal stated that Tecentriq + Carboplatin + Pemetrexed demonstrated favorable anti-tumor activity and a manageable safety profile in treatment-naive patients with advanced non-squamous NSCLC and brain metastases.
