Is Osimertinib Effective for Lung Cancer?

On June 8, 2023, Masahiro Tsuboi and colleagues from the National Cancer Center Hospital East published the results of the Phase 3 ADAURA clinical trial in "The New England Journal of Medicine." The study evaluated the efficacy and safety of adjuvant Osimertinib in patients with EGFR mutation-positive non-small cell lung cancer (NSCLC) after complete resection.

ADAURA is an international, randomized, double-blind Phase 3 trial. Patients with completely resected, EGFR mutation-positive NSCLC, regardless of prior adjuvant chemotherapy, were randomized 1:1 to either the Osimertinib group (80mg once daily for up to 3 years) or the placebo group. The primary endpoint was disease-free survival (DFS), and the secondary endpoint was overall survival (OS).

The trial included 682 participants, with 339 in the Osimertinib group and 343 in the placebo group. The 5-year OS rates for Stage II/III patients were 85% in the Osimertinib group versus 73% in the placebo group. For the overall population, the 5-year OS rates were 88% in the Osimertinib group versus 78% in the placebo group.

Based on the ADAURA trial results, Tsuboi stated that adjuvant Osimertinib significantly improved overall survival compared to placebo in patients with completely resected, EGFR mutation-positive NSCLC.
