(Lung Cancer) Long-Term Overall Survival Improved with Opdivo Monotherapy?

On August 14, 2019, Scott J. Antonia published in the medical journal The Lancet the pooled analysis results of previously treated non-small cell lung cancer (NSCLC) patients receiving anti-PD-1 antibody Opdivo (nivolumab) monotherapy.

This trial was a pooled analysis of previously treated NSCLC patients who received Opdivo monotherapy after at least two prior treatment regimens, with overall survival (OS) as the primary endpoint. The pooled analysis was based on patient data from the CheckMate-017, CheckMate-057, CheckMate-063, and CheckMate-003 trials, with all patients followed for at least four years.

The results showed that among all patients (N=664), the primary endpoint of 4-year overall survival was 14%. For patients with PD-L1 expression ≥1%, the 4-year OS was 19%, and for those with PD-L1 expression <1%, it was 11%.

In the CheckMate-017 trial, which compared the efficacy of Opdivo monotherapy versus Docetaxel, the 4-year overall survival rates were: Opdivo group 14% vs. Docetaxel group 5%.

Based on the pooled analysis results, Scott J. Antonia concluded: Previously treated NSCLC patients receiving anti-PD-1 antibody Opdivo monotherapy showed improved long-term overall survival, with superior efficacy compared to Docetaxel.
