(Solid Tumors) Is Enhertu Effective?

From June 2-6, 2023, Funda Meric-Bernstam and colleagues from Memorial Sloan Kettering Cancer Center presented efficacy and safety results of Enhertu treatment in HER2-positive, advanced solid tumor patients from the DPT02 Phase 2 clinical trial at ASCO 2023 in Chicago, USA.

The DPT02 Phase 2 clinical trial was an open-label trial conducted jointly at multiple facilities in the United States and Japan. HER2-positive, advanced solid tumor patients received Enhertu 5.4-6.4 mg/kg (every three weeks) until disease progression or adverse events occurred. The primary endpoint was objective response rate, with secondary endpoints including duration of response, disease control rate, progression-free survival, overall survival, and safety.

Results from 267 patients at a median follow-up of 9.7 months showed: primary endpoint objective response rate of 37.1%, secondary endpoint median duration of response of 11.8 months; in HER2-positive patients, objective response rate was 61.3% with median duration of response of 22.1 months.

Regarding safety, Grade 3+ adverse events occurred in 58.4% of patients, treatment discontinuation due to adverse events in 11.6%, and interstitial lung disease/pneumonitis in 6.7%.

Based on Phase 2 clinical trial results, Funda Meric-Bernstam and colleagues concluded: HER2-positive, advanced solid tumor patients treated with Enhertu demonstrated good response rates with controllable adverse effects. Enhertu has the potential to become a new treatment option for HER2-positive solid tumor patients.
