AstraZeneca announced that in an international, multi-center Phase 3 clinical trial involving patients with HER2-negative metastatic breast cancer carrying BRCA1 or BRCA2 mutations, the PARP inhibitor Olaparib met its primary endpoint. This demonstrates that the drug is effective not only for ovarian cancer but also for breast cancer. The trial is named OLYMPIAD, and further information can be found online.
The OLYMPIAD trial focused on patients with metastatic breast cancer who had received prior treatments, such as anthracyclines or taxanes, or as adjuvant therapy after surgery. This drug has also undergone clinical trials in Japan.
As mentioned, this Phase 3 trial compared patients receiving Olaparib (300 mg twice daily) with a physician-selected treatment group (receiving either capecitabine, eribulin, or vinorelbine). Eligibility criteria included prior treatment with anthracyclines or taxanes for metastatic breast cancer, or as adjuvant therapy, or endocrine therapy for hormone receptor-positive patients.
The primary endpoint was progression-free survival (PFS). Secondary endpoints included overall survival (OS), second progression-free survival (PFS2), and health-related quality of life (HRQoL). Crucially, the trial results showed superior outcomes for the Olaparib group. Detailed findings will be fully presented at upcoming medical conferences. Currently, AstraZeneca is also conducting Phase 3 trials targeting patients with non-metastatic breast cancer.
