On May 18, 2023, Stefania Napolitano et al. from Università degli Studi della Campania "Luigi Vanvitelli" published in JAMA Oncology the efficacy and safety results of panitumumab + Lonsurf (trifluridine/tipiracil) as third-line treatment in refractory RAS wild-type recurrent colorectal cancer patients in the VELO Phase 2 clinical trial.
The VELO Phase 2 clinical trial was an open-label, randomized trial that randomized refractory RAS wild-type recurrent colorectal cancer patients 1:1 into the panitumumab + Lonsurf group and the Lonsurf monotherapy group. The primary endpoint was progression-free survival.
Patient characteristics for the 62 participants enrolled: median age was 65 years (range 39–81) for the panitumumab + Lonsurf group and (range 32–82) for the Lonsurf group; male percentage was 61.3% (N=19) for the panitumumab + Lonsurf group and 54.8% (N=17) for the Lonsurf group.
Trial results: the primary endpoint median progression-free survival was: panitumumab + Lonsurf group 4.0 months / Lonsurf group 2.5 months. The 6-month progression-free survival rate was: panitumumab + Lonsurf group 38.5% / Lonsurf group 13.0%. The 12-month progression-free survival rate was: panitumumab + Lonsurf group 15.4% / Lonsurf group 0%.
Based on the VELO trial results, Stefania Napolitano et al. stated: In refractory RAS wild-type recurrent colorectal cancer patients, panitumumab + Lonsurf as third-line treatment improved progression-free survival compared to Lonsurf monotherapy.
