Gastric Cancer: Andecaliximab Plus Nivolumab Phase II Results

On January 17-19, 2019, at ASCO Gastrointestinal Cancers Congress, Dr. Manish A. Shah presented Phase II trial results evaluating andecaliximab (anti-MMP-9 monoclonal antibody) combined with nivolumab versus nivolumab alone in gastric and gastroesophageal junction adenocarcinoma.

Trial Design

Patient Population: Previously untreated unresectable/recurrent gastric and gastroesophageal junction adenocarcinoma

Treatment Arms:

• Andecaliximab + Nivolumab
• Nivolumab monotherapy

Primary Endpoints: Response rate, progression-free survival, overall survival

Key Results

Efficacy Comparison: No significant statistical difference between combination and monotherapy arms

Response Rate: Similar between andecaliximab + nivolumab and nivolumab alone

Progression-Free Survival: No improvement with combination

Overall Survival: No benefit with combination approach

Efficacy in Previously Treated Population

While combination therapy showed modest trends in previously treated, recurrent, and gastroesophageal junction patient subsets, differences did not achieve statistical significance.

Clinical Conclusions

Phase II data do not support additional benefit from andecaliximab addition to nivolumab in gastric cancer, with monotherapy remaining preferred approach.
