On March 10, 2023, Dr. Renata Ferrarotto and colleagues from The University of Texas MD Anderson Cancer Center published the efficacy and safety results of a Phase 2 clinical trial evaluating the combination of Avelumab and Axitinib in patients with recurrent or metastatic adenoid cystic carcinoma (ACC) in the Journal of Clinical Oncology.
Study Design: This was an open-label, single-arm Phase 2 clinical trial evaluating the combination of Avelumab (an anti-PD-L1 antibody) and Axitinib (a VEGFR inhibitor) in 40 patients with recurrent or metastatic ACC. The primary endpoint was the objective response rate (ORR). Secondary endpoints included progression-free survival (PFS), overall survival (OS), and safety.
Clinical Outcomes: In the evaluable population of 28 patients:
- Objective Response Rate (ORR): 18% (exceeding the primary target).
- 6-Month ORR: 14%.
- Median PFS: 7.3 months.
- 6-Month PFS Rate: 57%.
- Median Overall Survival (OS): 16.6 months.
Safety Profile: The most common adverse events reported by patients included fatigue (62%), hypertension (32%), and diarrhea (32%).
Conclusion: Based on the Phase 2 trial results, Dr. Renata Ferrarotto concluded that the combination of Avelumab and Axitinib demonstrated significant activity in recurrent or metastatic adenoid cystic carcinoma, with an ORR of 18% successfully meeting the study's primary objective.
