(Endometrial Cancer) Verzenio + Letrozole Effective?

On September 29, 2022, Panagiotis A. Konstantinopoulos et al. from Dana-Farber Cancer Institute published in the medical journal Journal of Clinical Oncology the efficacy and safety results of Verzenio (abemaciclib) plus Letrozole as treatment for patients with estrogen receptor-positive recurrent endometrial cancer in a Phase II clinical trial at Dana-Farber Cancer Institute.
This Phase II clinical trial enrolled 30 patients with estrogen receptor-positive recurrent endometrial cancer (N=30). They received Verzenio plus Letrozole therapy, with the primary endpoints being objective response rate (ORR) and 6-month progression-free survival rate (PFS).
The results showed that the primary endpoint ORR was 30%. The other primary endpoint, median progression-free survival, was 9.1 months. The 6-month progression-free survival rate was 55.6%, and the median duration of response was 7.4 months. Regarding safety, the most common Grade 3 or higher adverse events observed in the majority of patients were neutropenia (20%) and anemia (17%).
Based on the Phase II trial results, Panagiotis A. Konstantinopoulos et al. concluded: Patients with estrogen receptor-positive recurrent endometrial cancer treated with Verzenio plus Letrozole demonstrated durable and promising anti-tumor activity.
